The domain has a consistent structural motif with three antiparallel beta sheets packing against one alpha helix. In KCTD5, KCTD2, and KCTD17, this domain functions as a G protein binding domain (GBD) that interacts with Gbeta/gamma subunits. In combination with the BTB domain and Cullin3, these proteins form a pentameric E3 ubiquitin ligase that triggers ubiquitination and degradation of Gbeta/gamma subunits, regulating G protein-coupled receptor (GPCR) signaling. The domain adopts different interfaces in different KCTD subfamily members, resulting in distinct substrate binding mechanisms. The domain forms part of a pentameric assembly in native KCTD proteins.

In KCTD1 (Swiss:Q719H9) this domain functions as a transcriptional repressor [2]. It also mediates homomultimerisation of KCTD1 and interaction of KCTD1 with the transcription factor AP-2-alpha [2-3].
