DescriptionGonorrhea, caused by Neisseria gonorrhoeae, presents a significant public health challenge due to increasing antimicrobial resistance.[A275268] Zoliflodacin is a first-in-class spiropyrimidinetrione antibiotic designed to address this need by inhibiting bacterial type II topoisomerases (DNA gyrase and topoisomerase IV), thereby blocking bacterial DNA synthesis.[A274938,L54748] It is indicated as a single-dose oral therapy for the treatment of uncomplicated urogenital gonorrhea.[L54748] Developed through a collaboration involving the Global Antibiotic Research and Development Partnership (GARDP), zoliflodacin received approval from the U.S. FDA in December 2025, following Fast Track, Qualified Infectious Disease Product, and Priority Review designations.[L54743,L54803]
