Separin is a cysteine protease critical for chromosome segregation during mitosis and meiosis, exhibits a conserved multi-domain architecture across eukaryotes. Separin contains an N-terminal HEAT/ARM repeats which allows the protein to bind regulatory partners like securin and cyclin B1; followed by a TPR-like domain which binds phosphorylated residues and a C-terminal caspase-like protease domain which contains a catalytic triad (Cys-His-Asp) and recognises substrates with a conserved arginine at the P1 position [1,3]. The C-terminal domain is structurally related to caspases but includes a unique substrate-binding subdomain (PPD) [2,4].
