This entry represents the Fibronectin type III (FN3) domain found at the N-terminal of Interleukin-27 subunit beta (IL27B), from animals. This domain may be found in association with a C-terminal FN3 (Pfam:PF00041). Also known as EBI3, IL27B binds p28 to form the heterodimeric cytokine IL-27, which plays an important role in innate immunity by inducing naive CD4+ T cells proliferation [1]. IL-27 binds GP130 and IL-27Ra, acting as a bridge, to activate the JAK/STAT signaling cascade. EBI3 stabilizes the complex by binding a positively charged surface of IL-27Ra and Domain 1 of GP130 [2].

This entry describes the domain 4 in Leukemia inhibitory factor regulator (LIFR) and other type I cytokine receptors found in mammals. They are involved in signal transduction for various cytokines, playing crucial roles in immune response, hematopoiesis, and development. LIFR extracellular region is composed of D1-D2 (Pfam:PF17971) which is a cytokine binding homology region (CHR) followed by an Ig-like domain D3 (Pfam:PF21177), another CHR region composed of D4 and D5 [1-2] and three membrane-proximal fibronectin type-III (FNIII) domains (Pfam:PF00041).
