Comparison of the refined model with the recently reported X-ray crystallographic structure of the trimeric complex of S. pombe Uba1-Ub-Ubc4 (PDB ID: 4II2) lends support to the theoretical 3D model of... the quaternary complex.

Steered Molecular Dynamics and refinement of the final complex Comparison of the final MD structures and the recently solved X-ray structure of Uba1 in complex with Ubc4 (PDB entry 4II2; [14] ) showed that the loop masking the hUbA1 catalytic cysteine (Cys-cap loop) prevented a close packing between UbcH10 and the ternary complex.

In contrast, in the available structure of the E1–E2 complex (PDB structure 4II2) the Cys-cap loop is not assigned, thus suggesting a large flexibility in the covalent construct that mimics the thioester crosslinking event.
