To better understand the binding mode and how this ligand interacts with the glycine-rich loop, we crystallized 1b in complex with wild type p38α and were surprised to find the molecule bound exclusively within the p38α MAPK insert ( Figure 1A ) (PDB code 4DLI), far away from the glycine-rich loop.
