Bcl-xL bound to ABT-737 (PDB ID 2yxj), a compound similar to ABT-737 (PDB ID 3qkd), an unrelated Bcl-xL inhibitor (PDB ID 3zln), and Mcl-1 bound to an Mcl-1 inhibitor (PDB ID 4hw2) were rotated for 10... random orientations, and an all-vs-all comparison of the ensemble of ABT-737 bound exemplars to those generated for each orientation was performed.

Not surprisingly, we found that the members of the experimental structure ensemble that scored better than the cosolvent aMD were the structures from the Lessene hit co-complex itself (PDB codes: 3ZK6... 3ZLN, 3ZLO, 3ZLR).
We note that several co-crystal structures with Bcl-xL have been determined for various derivatives of the original HTS hit (PDB codes: 3ZK6, 3ZLN, 3ZLO, 3ZLR).
