Superposition of the crystal structures of the triple TPR tandem of Bub1 (PDB 3ESL) and BUBR1 (PDB 2WVI) with the TPR1 domain of HOP in complex with a heat shock cognate 70 (HSC70) peptide (PDB 1ELW);... the TPR2A domain of HOP in complex with a HSP90 peptide (PDB 1elr)  ; the PP5 TPR in complex with a HSP90 peptide (PDB 2BUG)  and the PEX5 TPR in complex with a peroxisomal targeting signal-1 (PTS1) peptide (PDB 1FCH)  show that BUB1 and BUBR1 residues involved in binding blinkin are mapped onto the TPR-forming helices that are involved in protein–peptide interactions ( Figure 2 B).

Therefore, the similar topology suggests that BUBR1 and Blinkin may interact in a mode similar to that observed in complexes formed between TPR PP5 and a Hsp90 peptide (PDB 2BU... ) ( Cliff et al., 2006 ); TPR HOP-Hsc70 peptide (PDB 1ELW) ( Scheufler et al., 2000 ); TPR HOP-Hsp90 peptide (PDB 1ELR) ( Scheufler et al., 2000 ); and TPR PEX5 in complex with a peroxisomal targeting signal-1 (PTS1) peptide (PDB 1FCH) ( Gatto et al., 2000 ).
