Inspection of the yeast RNAPII X-ray structure of an elongation complex ( 7 ; PDB #1SFO) suggests that an evolutionary invariant residue ( sc RPB1-G835 in yeast RNAPII, orthologous to mj A′-G8... 5 in M. jannaschi ; Figure 4 a) may have been specifically selected for the small size of its side chain to allow an unimpeded passage of the DNA template strand into the active site.
Note the close spatial proximity between the G825 side chain and the phosphodiester backbone of the DNA [data from PDB #1SFO ( 5 ) visualized with Cn3D; www.ncbi.nlm.nih.gov/Structure/CN3D/cn3d.shtml ].
